- A large room is depicted which the artist had seen again and again in Tornabuoni's house, with the walls divided by pilasters carved with elaborate candelabras which are repeated, perhaps on leather but more probably on wood or pietra serena in all the panels of the background.
- At the entrance stairway a group of visiting women have just entered.
The women had been placed in the scene to serve as public spectators, detached from any action like devotees at the threshold of a temple.
- Standing out from the others in profile is a young woman, almost a girl, wearing gold embroidered brocade, possibly for the first time dressed like a lady and a little embarassed by the stiffness of the garment and its large, rather long sleeves.
She may be Giovanna Tornabuoni, the banker's only daughter, here acting out her first official role at the ceremony in place of her dead mother.
Behind her are four older women, uninhibited in their postures and gestures.
- The most important figure, however, is the young maiden who provides a contrast with the figure of the old St Anne in her large bed.
The wet nurses busy themselves with preparing the bath of the soft, pink, newborn baby, which seems to respond to the tender smile of the young woman holding the infant in her arms.
